Dulbecco's Modified Eagle Medium (DMEM) High Glucose without L-glutamine is a widely used standard cell culture medium. It provides basic nutrients for the growth of a variety of mammalian cell lines, especially when supplemented with Fetal Bovine Serum. In the past, DMEM was shown to successfully cultivate primary cells, such as fibroblasts, HUVECs, neurons, smooth muscle cells, or even cell lines, such as HEK-293, HeLa, Cos-7, and PC-12. DMEM comes in various formulations, in which the composition of components may differ.
The original formulation of DMEM contains 1.0 g/L. In contrast, DMEM High Glucose has an increased glucose content of 4.5 g/L, and does not contain L-glutamine.
DMEM is based on Eagle's Minimal Essential Medium (EMEM) but has a fourfold higher concentration of amino acids and vitamins.
A widespread practice is the supplementation of DMEM with FBS, typically at a concentration of 10%, and this is often necessary as the medium does not contain any growth hormones, lipids, or proteins essential for cell growth.
DMEM contains a buffer to maintain pH levels and usually comes with phenol red as a pH indicator. Cells cultivated in DMEM or any other medium containing the bicarbonate buffer system must be kept in a controlled CO2 environment of 5-10%. With the added phenol red, pH changes from 6.2 (yellow) to 8.2 (red) can be monitored.
