Biotin binds to the binding pockets of streptavidin, Strep-Tactin®, and Strep-Tactin®XT with distinct affinities. It can be used for the preparation of Strep-Tactin®XT elution buffer, Buffer BXT.
When biotin is applied to Strep-Tactin®XT resin, it displaces bound Strep-tag®II or Twin-Strep-tag® proteins, allowing an efficient elution. It can also be used for eluting biotinylated proteins from Strep-Tactin® resins. It should be noted that Strep-Tactin®resins cannot be regenerated after biotin application.
